The Adelaide Football Club was the first AFL contingent to board the aircraft, leaving Adelaide on Monday for a training camp on the Gold Coast.
The jet features the AFL logo, as well as the logos of each of the 17 AFL clubs.
Virgin Blue Group of Airlines chief executive commercial Liz Savage said Virgin Blue was proud to have come on board as the new official airline of the AFL.
"This is just one of the many ways that we will support the AFL in our new partnership and we look forward to providing the AFL and the players with great service on our extensive network," Savage said.
The AFL-branded aircraft will carry Virgin Blue passengers, AFL players and supporters all around Australia.
AFL general manager of commercial operations Darren Birch said fans would get a kick out of flying on the one-of-a-kind jet.
"The fact that the design incorporates all 17 AFL club logos is a great show of support for the game and for the individual clubs," Mr Birch said.
"Virgin Blue has been working closely with us to ensure that players will be well accommodated and we are very impressed with the way the airline is working its way through each club's arrangements and their specific requirements."
