The 26-year-old Roughead is now seven goals ahead of Collingwood's Travis Cloke, with the Magpies facing North Melbourne at the MCG on Sunday.
GWS Giants star Jeremy Cameron and West Coast Eagle Josh Kennedy are one further back on 60 goals.
The Giants are away to the Gold Coast Suns in round 23, while the Eagles host the Adelaide at Paterson Stadium on Saturday night.
All four players have never won the Coleman Medal.
Roughead, who kicked one goal in the first, one in the third and two in the fourth on Friday night, said he has enjoyed an extended time in the forward line this year, although he hasn't really focused too much on the race for the Coleman.
"I don't think we played that kind of stuff," Roughead said after the match.
Aside from Roughead, the Hawks have three other players who have kicked more than 30 goals: Lance Franklin (58), Luke Breust (37) and Jack Gunston (35).
"If we were going to get better, we were going to have to have different avenues towards goal," Roughead said.
"This year we probably had three or four blokes that have kicked over 30 goals, which is always good going into finals because defences don't really know how to match up against us."
Hawthorn coach Alastair Clarkson said a Coleman Medal would be just reward for Roughead after an outstanding year, but added there were other more important goals for his side after it wrapped up the minor premiership for the second successive season.
"If he doesn't (win the award), we've got bigger things hopefully in the next few weeks," Clarkson said.
Reigning Coleman Medallist Jack Riewoldt is likely to be too far back to claim a third medal, starting the final round of the season on 57 goals ahead of Richmond’s meeting with Essendon on Saturday night.
