Daniel Bradshaw (hamstring), Travis Johnstone (hamstring) and Justin Sherman (back) all ran at top speed at various stages of the 90-minute hit-out.
The trio missed the Lions' 13-point win over Adelaide last weekend, and while not confirmed starters yet, look likely to be added to the squad.
The match against Melbourne is the first of four successive fixtures against teams that are lower on the table than the Lions.
Defender Joel Patfull said having the injured players available would be a huge boost, particularly Bradshaw, who had booted 43 goals before injuring his hamstring in round 11 against Fremantle.
"On the weekend we got it inside 50 a fair bit but we didn't convert as much as we could and I think Braddy (Bradshaw) will definitely help with that," he said.
"We've got a pretty good run the next few weeks but they're all must-win games, we're sitting in sixth now so a few good wins will set us up pretty well for September.
"We had a bit of a mishap against the Bulldogs, they beat us all over the ground really, we didn't win the contested footy, but it's something we've worked on and I think on the weekend we took a good step forward.
"They've (Melbourne) been playing some decent footy, they've only won the one game, but them in Melbourne, they always play pretty well at the MCG so we'll have to be on our game."
Patfull is playing his 50th game for the Lions on the weekend after crossing from Port Adelaide at the start of 2006.
He has become part of a consistent and underrated back six that includes Daniel Merrett, Joel Macdonald and Troy Selwood.
"We've played together for a while and we're getting a good bond back there," he said.
"Just getting the games together and playing as a group has really helped."
