The Saints jumped to a 20-point half-time lead and, despite a Todd Banfield led revival by the Lions early in the second half, ran out comfortable winners.
The young Brisbane Lions line-up, coming off a 62-point thrashing to Essendon in the opening match of the night, received a boost from experienced inclusions Daniel Merrett, Ash McGrath and Joel Patfull.
But despite showing improved endeavour, the Lions were again unable to do any damage on the scoreboard in the first half.
In a scrappy opening, the match went back-and-forth for the first five minutes before Brett Peake got the Saints on their way with a running supergoal.
Two more first half goals to the Saints followed, both a result of Lions errors.
First, Alistair Smith was collected late after marking in the centre square and converted after receiving a 50m penalty. Then, Tom Lynch was gifted an easy goal thanks to a Lions turnover deep in defence.
Brisbane finally gathered some momentum early in the second half, with two goals to Banfield closing the margin to nine points.
But former Cat Gamble ended hopes of a Lions comeback with an electrifying burst in front of goal.
The key forward kicked two near-identical snaps on his non-preferred left foot in quick succession, before adding a third at the 17-minute mark courtesy of a Brendon Goddard handball to put the game away.
Goddard was a clear best on ground for St Kilda, collecting 11 disposals and using the ball superbly, while Nick Dal Santo gathered 12 possessions in a typically composed performance.
For the Lions, Daniel Rich recovered from a disappointing outing against the Bombers, gathering 13 touches through the midfield, while Banfield's two goals and flashing centre square runs provided a spark missing for much of the Lions' night.
