Youngsters Jack Ziebell and Liam Anthony will play their first games for the club as will second-year rookie and recent Indigenous All-Stars representative Cruize Garlett.
"Ziebell has been going pretty well in the last three or four months and he'll be given an opportunity tomorrow night, which will be great for our supporters," coach Dean Laidley said on Thursday at Arden Street.
"Liam Anthony and Cruize Garlett will definitely play their first games and I think that is great.
"Cruize has been here a year, he's still a rookie and played in the All-Star game a couple of weeks ago, and I thought he was pretty good.
"And Liam will also play his first game, which is great for us."
Laidley said Garlett has come into contention after spending a year playing in North Ballarat's VFL reserves team and working hard on his fitness.
"Twelve months ago, he was running laps around the boundary line and he was carrying a bit of a gut," he said.
"Now, 12 months on, he's improved dramatically."
Talented forward Aaron Edwards will not figure in the match after an off-field indiscretion saw him suspended for all of the NAB Cup and until round five of the regular season.
Laidley said Edwards, who booted four goals in the last quarter of the Roos' recent intra-club match, should be fully fit when he returns.
"He'll be playing in all the practice games and at VFL level, and the VFL doesn't start until round three for us so he's going to be playing practice games right up to there," he said.
"In essence, he might miss a week where he's not playing a game of footy. He's probably going to play five or six or even seven practice games leading up to that, so I wouldn't have thought it's going to be a big issue with his conditioning."
Young gun Jesse Smith will also sit out Friday night's match but will play the following week.
However, emerging ruck prospect Todd Goldstein is expected to play, with Laidley confident there is room for him alongside fellow big men David Hale, Drew Petrie and Hamish McIntosh.
