The Collingwood full-forward travelled to Brisbane with the team but contracted a virus that ruled him out.
He has been replaced in the selected side by Marty Clarke.
“Unfortunately Trav has had a little bit of gastro overnight," Collingwood assistant coach Tarkyn Lockyer told Croc Media.
"We were hoping he’d come up, but unfortunately he’s pretty lethargic and has pulled out of the side, which gives Marty Clarke an opportunity.
"We gave him obviously plenty of time this morning to try to get it right, he trained last night and was ok - it’s just one of those things that happens.
"It does mean that we will have to change our forward line marginally," Lockyer conceded.
"He’s such a strong presence up there and obviously someone we kick the ball to a fair bit, but we’re more than capable at ground level as well.
"We’ve got a number of other guys who are strong overhead with the likes of Jamie Elliott and Andrew Krakouer, as well as obviously Quinten Lynch.”
Cloke's absence is a huge blow for the Magpies (5-4) who desperately need to win to move back towards the top eight.
Heath Shaw has returned to the team after overcoming a hamstring injury, as has Alan Didak who will start as the sub.
Cloke was set to cause match-up nightmares for the Lions whose defence has been depleted by the suspension of Daniel Merrett and the long-term leg injury to Matt Maguire.
Sam Docherty is the Lions' sub.
